The Ghana international has been out of first-team football for six weeks, after injuring his hamstring earlier this year in an English Premier League game against Bournemouth.
The 23-year-old returned to first-team training last week but is still far off full match sharpness, hence the decision by club manager Claude Puel to draft him into the Under-23 setup.
He is expected to start for the side when they take on the Aston Villa U-23s on Friday night.
Amartey has made eight appearances so far this season for the foxes.
